GATA3 Expression and Relationship between Clinicopathological Parameters in Invasive Breast Carcinomas

Background: GATA3 is a transcription factor which regulates luminal cell differentiation in breast tissue and its expression is associated with estrogen receptor (ER) gene.
Design: We immunohistochemically stained GATA3, CK5/6, EGFR, CK14 and vimentin on tissue microarray blocks of 457 invasive breast carcinomas. Tumor are subclassified as Luminal A, Luminal B, HER2 expressing, basal-like and null type according to ER, PR, cerbB2, CK5/6 and EGFR expressions. 13-84 months follow-up data for 254 cases were obtained. Association between GATA3 expression with clinicopathological parameters, CK5/6, CK14, EGFR, vimentin expression, molecular subtypes, disease free survival (DFS) and overall survival (OS) were evaluated.
Results: 215/457 (47%) tumors were GATA3 positive. GATA3 expression was inversely correlated with mitotic count (p<0.0001), nuclear grade (p=0.001), histological grade (p=0.001), tumor necrosis (p=0.001), nipple invasion (p=0.01), metastasis (p=0.03), vimentin (p=0.0003), EGFR (p=0.015) and CK14 (p=0.001) expressions; and directly associated with ER (p<0.0001) and PR (p<0.0001) expressions. No relationship can be showed between patients age, breast skin and fasia invasion, tumor size, axillary lymph node metastasis, cerbB2, CK5/6 expression and GATA3. The highest GATA3 expression was in luminal A type (140/245) and the lowest expression was in basal-like (1/42), (p<0.0001). Only 1 patient (1/15, 6.6%) died of breast carcinoma had GATA3 expression (p=0.001). GATA3 positivity was associated with good DFS and OS (p=0.001 and p=0.0009).
Conclusions: These results showed that (1) ER and GATA3 expression have strong correlation, (2) GATA3 expression is lost in mitotically active, high grade breast carcinomas, (3) LuminalA breast carcinomas have the highest GATA3 expression while basal-like carcinomas have the lowest and (4) GATA3 positivity can be used as a prognostic factor to predict good DFS and OS.
